Cover Story: Precision medicine is based on the observation of major inter-individual variability in drug responses. Drug metabolic enzymes and transporters are responsible for important variability in drug disposition due to genetic polymorphisms or environmental factors such as dietary components, toxins, or drugs. Thus, in vivo evaluation of the activity of these enzymes and transporters is of great importance. A cocktail approach involving the administration of multiple specific probe drugs can be used to simultaneously assess their activity. The aim of this work is to evaluate potential drug–drug interactions between probe drugs of the Geneva cocktail, with a focus on the pharmacokinetic interaction between cytochrome P450 probe drugs and fexofenadine (P-glycoprotein substrate).
