Cover Story: The plague bacillus Yersinia pestis relies on metabolic adaptation to survive and persist within its flea vector. Here, we investigated the respective roles of transketolase (TktA) and transaldolase (TalB), two key enzymes of the non-oxidative pentose phosphate pathway. We found that TktA is essential for growth and metabolic homeostasis, whereas TalB specifically promotes biofilm formation and the sustained colonization of the flea foregut. Metabolomic analyses revealed that these enzymes play distinct and non-redundant roles in coordinating carbon flux distribution, nucleotide metabolism, and redox balance. Our findings identify the non-oxidative pentose phosphate pathway as a central metabolic hub linking bacterial physiology to flea colonization dynamics. View this paper
