Cover Story: Peripheral artery disease (PAD) reduces blood flow and oxygen delivery to skeletal muscles, leading to ischemic injury and impaired tissue repair. We compared age-dependent responses to hind limb ischemia in young and old mice using multispectral optoacoustic tomography (MSOT), laser speckle contrast imaging (LSCI) and histology. MSOT and LSCI demonstrated reduced oxygenation and perfusion following femoral artery occlusion, with progressive recovery which was delayed in old mice. Histology showed that young mice exhibited better tissue repair, characterized by increased vascularization and cell proliferation with reduced fibrosis. These findings highlight age-related differences in vascular and tissue remodeling following ischemic injury and support the use of non-invasive imaging for longitudinal assessment of PAD. View this paper
